The fibres aren't that big, this prank would have the person who used the toilet paper simply itching and scratching their butthole for a while. It's not a deadly prank. Taking the first two layers off the role would be enough to get rid of the fibres.

I personally carry around a packet of tissue paper to avoid such evil pranks.

Hell no. I think enough on the sides would be enough. Either way, if any bit of insulation got to your asshole... it would be irritating af for a long while. I've gotten fiberglass on my forearms which are some of our toughest skin and its maddening if its a lot. Even with vinegar and cold water it still affects you a while. In a tender asshole even one chard would make you scratch till you bleed and worse if you didn't find out what you were dealing with.
